Endothelial Differentiation Gene-1, a New Downstream Gene Is Involved in RTEF-1 Induced Angiogenesis in Endothelial Cells

Abstract: Related Transcriptional Enhancer Factor-1 (RTEF-1) has been suggested to induce angiogenesis through regulating target genes. Whether RTEF-1 has a direct role in angiogenesis and what specific genes are involved in RTEF-1 driven angiogenisis have not been elucidated. We found that over-expressing RTEF-1 in Human dermal microvascular endothelial cells-1 (HMEC-1) significantly increased endothelial cell aggregation, growth and migration while the processes were inhibited by siRNA of RTEF-1. In addition, we obser… Show more

“…Above all, TEF3-1 is important for physiologically processes such as cell proliferation, cell migration, mammalian cell fate specification [32][33][34][35]. Recent paper mentioned that TEF3-1 is a prognostic marker for cancer progression and a potential target for cancer treatment [33,[36][37][38].…”

“…Matrigel assay was performed as described previously. 49 Briefly, bEnd3 endothelial cells were plated in a six-well Matrigel plate (BD Biocoat Matrigel Matrix, 354432). The cells were seeded at a density of 10 5 cells per well in 2 mL medium.…”

“…Cells were transfected with Lipofectamine 2000 (Invitrogen) according to the manufacturer's suggestions. Human dermal microvascular endothelial cells (HMEC-1) and Human umbilical vein endothelial cells (HUVEC) [ 30 , 31 ], which have endothelial characteristics of cell migration and tubular structure formation, were purchased from Science Research Laboratories (ScienCell Research Laboratories, San Diego, CA, USA) and stored by the lab of Institute of Biomedical Sciences and School of Life Sciences, East China Normal University. HMEC-1 cells were maintained in MCDB-131 medium (Invitrogen, Life Technologies) supplemented with 10% fetal bovine serum (FBS), 10 ng/mL of human epidermal growth factor (EGF) (Sigma, St. Louis, MO), 1 μg/mL of hydrocortisone, 5 mg/mL of L-glutamine, and antibiotics (penicillin-streptomycin) at 37°C with 5% CO 2 .…”
