Endothelial repair in stented arteries is accelerated by inhibition of Rho-associated protein kinase

Abstract: AimsStent deployment causes endothelial cells (EC) denudation, which promotes in-stent restenosis and thrombosis. Thus endothelial regrowth in stented arteries is an important therapeutic goal. Stent struts modify local hemodynamics, however the effects of flow perturbation on EC injury and repair are incompletely understood. By studying the effects of stent struts on flow and EC migration, we identified an intervention that promotes endothelial repair in stented arteries. Methods and ResultsIn vitro and in viv… Show more

“…Comprehensive reviews already exist, in which these techniques are described in detail . In brief, systems range from common planar flow chambers, in which shear stress is applied via the laminar flow of fluid between two parallel plates, to simple circular channels, microvascular networks, and patient‐specific geometries.…”

“…PTA is a therapeutic procedure alleviating the symptoms in a number of atherosclerotic conditions [20]. Its effectiveness in both coronary artery disease and peripheral arterial disease is, however, limited by the process of remodelling leading to intimal proliferation and recurrence of vascular occlusion [2123].…”

“…Frequency (Δf) and dissipation (ΔD) signals have been detected during drug injection and static incubation for about 1 h at 37 °C. Two different types of cytoskeleton drugs were used: i) CytoD, that hinders the polymerization of actin in the cytoskeleton and induces dramatic changes in cell morphology, and ii) Y27632 that performs pharmacological inhibition of Rho‐associated protein kinase activity …”
