Enhanced Diffractivity of Phosphoglucomutase Crystals. Use of an Alternative Cryocrystallographic Procedure

Abstract: A continuous procedure for replacing the 2M (NH4)2SO4 within crystals of rabbit muscle phosphoglucomutase by 55% polyethylene glycol 600 (PEG 600) is described. The success rate (absence of fracturing) approaches 100% in spite of the fragility of the crystals. The procedure is based on the use of a biphasic PEG/salt mixture in conjunction with a flow cell that allows several crystals to be treated at once.
