Enhanced Replication of Hepatitis E Virus Strain 47832c in an A549-Derived Subclonal Cell Line

Abstract: Hepatitis E virus (HEV) is a human pathogen with increasing importance. The lack of efficient cell culture systems hampers systematic studies on its replication cycle, virus neutralization and inactivation. Here, several cell lines were inoculated with the HEV genotype 3c strain 47832c, previously isolated from a chronically infected transplant patient. At 14 days after inoculation the highest HEV genome copy numbers were found in A549 cells, followed by PLC/PRF/5 cells, whereas HepG2/C3A, Huh-7 Lunet BLR and … Show more

“…Recently, successful replication of HEV-3c strain 47832c (GenBank accession No. KC618403), in A549/D3 cells was demonstrated by Johne et al (2016;Schemmerer et al, 2016).…”

“…4) while MeT's, derived from Sar-55, Mex-14, ZJ-1 or Kernow-C1, had little, if any, effect. It is even more interesting because HEV 47832c strain is a cell culture-adapted strain (Schemmerer et al, 2016). A simple hypothesis would be that HEV adaption to cell culture requires effective blockade of the IFN-β signaling.…”

“…In order to determine the limit of quantification of the infectivity assay, cell culture-adapted HEV-3 strain 47832c at ten concentrations from 5×10 2 to 1×10 6 genome copies per well were used to infect A549/D3 cells in triplicate experiments (29, 30). The infected and control cells, which were not exposed to viral particles, were cultured for 14 days.…”

“…However, due to the lack of reliable infectivity assays, most HEV inactivation studies to date have been limited to using surrogate viruses (27, 28). Recently, successful replication of HEV-3c strain 47832c (GenBank accession # KC618403), in A549/D3 cells was demonstrated by Johne and coworkers (29, 30). This system has been employed to study the temperature sensitivity of HEV (30), demonstrating a potential for this system to be used in other HEV inactivation studies.…”
