Enrichment procedures for the isolation of Salmonella, Arizona, Edwardsiella and Shigella from faeces

Abstract: SUMMARYStrontium selenite A broth incubated at 370 C., and strontium chloride B broth incubated at 430 C., have been found an effective combination for the isolation of Salmonella, Arizona and Edwardsiella from human faeces.Direct plating on deoxycholate citrate agar was superior to enrichment methods for the isolation of Shigella species.

“…We have previously evaluated several liquid media for enrichment (unpublished data). These were strontium hydrogen selenite and strontium chloride B enrichment broth (Iveson, 1973), trypticase soy broth and tetrathionate broth (Bailey and Scott, 1974), potassium tellurite and alkaline peptone water (Furniss, Lee and Donovan, 1978), and nutrient broth (Cowan, 1974). Nutrient broth was the most satisfactory and produced excellent enrichment after incubation of faecal samples for 24 h. In a large epidemiological study of children with and without diarrhoea we found that Aeromonas spp.…”

“…is that protein which is carried over from the non-selective enrichment broth exerts a negative effect upon the selectivity of the bi-selenite ion (Leifson, 1936;North and Bartram, 1953). Incubation of the selective enrichment broths at 43° rather than at 37° may have reduced the selectivity of strontium bi-selenite, but this effect was not apparent in the results of Iveson (1973). The effect of alteration from 0-1% w/v potassium di-hydrogen phosphate in the formulation of S.S.B.…”
