A espécie Erythroxylum, conhecida como 'coca', composta por diversos alcalóides, tem a cocaína como seu principal componente ativo. A folha de coca é reconhecida e utilizada há centenas de anos para uma ampla gama de propósitos, desde aplicações terapêuticas até usos recreativos. Desde o século XX, a cocaína é considerada uma substância ilícita, mas por décadas, a folha de coca tem sido empregada pelos povos andinos para o tratamento e alívio de enfermidades como os males da altitude e como estimulante. Estudos iniciados na Europa no século XIX sobre sua eficácia como anestésico local em cirurgias oftalmológicas e odontológicas foram muito populares até que se descobriu seu potencial para causar toxicidade e dependência. Desde essa descoberta, 383 compostos diferentes já foram identificados em várias espécies de Erythroxylum, incluindo diterpenos, triterpenos, flavonoides, alcaloides e outros derivados. Apesar da complexidade química da planta, os potenciais terapêuticos da cocaína ainda carecem de uma exploração científica abrangente. Em contraste, seu uso farmacêutico, especialmente no sistema nervoso, atuando como anestésico local e antidepressivo, é bem estabelecido. Este estudo visa fornecer informações relacionadas ao gênero Erythroxylum, abordando suas atividades farmacocinéticas e farmacodinâmicas no contexto do uso medicinal contemporâneo, incentivando pesquisas para um entendimento mais profundo deste composto ativo.

Erythroxylum P. Browne is the largest and most representative genus of Erythroxylaceae family. It contains approximately 230 species that are mainly distributed in tropical and subtropical regions. Some species in this genus, such as E.monogynum and E.coca, have been used as folk medicines in India or South America for a long history. It is well known that Erythroxylum plants are rich in tropane alkaloids, and the representative member cocaine shows remarkable activity in human central nervous system. However, many other types of active compounds have also been found in Erythroxylum along with the broadening and deepening of phytochemical research. To date, a total of 383 compounds from Erythroxylum have been reported, among which only 186 tropane alkaloids have been reviewed in 2010. In this review, we summarized all remained 197 compounds characterized from 53 Erythroxylum species from 1960 to 2021, which include diterpenes, triterpenes, alkaloids, flavonoids, and other derivates, providing a comprehensive overview of phytoconstituents profile of Erythroxylum plants. In addition, the biological activities of representative phytochemicals and crude extracts were also highlighted. Graphical Abstract

Tropane alkaloids are bioactive metabolites with great importance in the pharmaceutical industry and the most important class of natural products found in the Erythroxylum genus. However, these compounds are usually separated by traditional chromatographic techniques, in which the sample is progressively purified in multiple chromatographic steps, resulting in a time- and solvent-consuming procedure. In this work we present the isolation of a novel alkaloid, 6β,7β-dibenzoyloxytropan-3α-ol, together with the two known 3α-benzoyloxynortropan-6β-ol and 3α,6β-dibenzoyloxytropane alkaloids, directly from the crude alkaloid fraction from the leaves of Erythroxylum subsessile, by using a single run pH-zone-refining counter-current chromatography method. The ethyl acetate/water (1:1, v/v) biphasic solvent system with triethylamine and HCl as retention and eluter agents, respectively, was used to isolate tropane alkaloids for the first time. The structures of the isolated alkaloids were elucidated by spectroscopic methods.
