The effects of environmental factors (photoperiod and light intensity) on in vitro tuberization were analysed on five potato cultivars of different genetic origin representing various maturity groups. No growth regulators were added to the culture media to avoid the potential effect(s) of growth regulators on the response to environmental stimuli. An 8% sucrose treatment was used for tuber initiation. Light (short-day treatment) applied after the induction phase delayed or inhibited tuber initiation. By contrast, darkness applied after the tuber induction stage accelerated and synchronized tuber initiation after high light intensity. No relationship was observed between the maturity groups of the tested cultivars and their tuber initiation response. The tuber number (.2 mm) per shoot varied from 1.19 to 1.52 depending on the cultivar in the best treatments. Consequently, the manipulation of light alone gave reliable tuberization.

Resume.-Pour faciliter l'etude des differentes etapes de Ia croissance et de Ia tuberisation chez Ia pomme de terre, un modele de developpement synchrone a ete mis au point in vitro a partir de fragments uninodaux de tiges. Pour y parvenir, seuls les facteurs de l'environnement physique et nutritionnel ont ete modifies, en absence de tout regulateur de croissance exogene pouvant perturber les teneurs ou activites de substances endogenes, recherchees par Ia suite comme marqueurs des etapes de Ia tuberisation. Le preconditionnement des plantes-mere, le niveau d'origine de I' explant, les photo-et thermoperiodes et les teneurs en azote et en sucre des milieux de culture sont des facteurs essentiels du conditionnement permettant d'optimiser et de synchroniser Ia tuberisation de Ia pomme de terre in vitro.Summary.-In order to study the different steps of the growth and of the tuberization process in potato, a synchronous development model was achieved in vitro from uninodal stem fragments. This result was obtained by only modifying the physical and nutritional factors of the environment. No growth regulator was added to any culture medium in order to avoid any modification in the content level or activity of endogenous substances which could be further analysed as key factors in the tuberization process. The mother plant preconditioning, the origin level of the explant, the photo and thermoperiod as the nitrogen and sugar content of the culture medium are essential factors to take into account in order to optimize and synchronize the in vitro tuberization process.

In vitro tuberization responses of eleven clones of potato were evaluated under different photoperiods and light intensities. No growth regulators were added to the medium to avoid the potential modification of the response to the environmental stimuli.Plantlets were grown on MS medium containing 3% sucrose for four weeks under long days (16 h). Tuberization was induced by short days (8 h). Five photoperiodic combinations and three light intensity treatments were applied.Light applied after the tuber induction phase delayed or inhibited tuber initiation (at proper photoperiods both at 111 and 55 ~mol m 2 s 1 light intensities at 24/15 ~ day/night temperature). Darkness following the induction stage accelerated and synchronized tuber initiation after high light intensity. However, these effects of dark treatment depended on light intensity applied in short days before dark treatment. There was an interaction between the duration of darkness applied in photoperiodic treatments and the light intensity applied in short days on tuber initiation. No relationship was observed between the maturity groups of clones tested and their tuber initiation responses. Tight interaction (P<0.01) of treatments and clones was proved by statistical analysis. Reliable tuberization and high rate of tuberization (1.20-1.52) can be reached using these hormon-free systems.
