“…MC3-R-and MC4-R-expressing sublines were identified by screening 8 -12 colonies for specific binding of [ 125 I]-NDP-MSH. In cell sublines selected for additional study, the presence of functional MCR having agonist binding profiles and coupling to adenylate cyclase consistent with those reported previously Alvaro et al, 1996) (Sawyer et al, 1980) was prepared by modifications of methods published previously (Tatro and Reichlin, 1987;Tatro, 1993), using a standard method (Thorell and Johansson, 1971) for lactoperoxidase-catalyzed radioiodination. Briefly, 4 g of NDP-MSH in 100 l of 10 mM sodium phosphate, pH 6.5, was added to 10 l (1 mCi) of [ 125 I]-Na (Amersham, Arlington Heights, IL) and 5 l of lactoperoxidase (0.8 mg/ml) (Calbiochem, La Jolla, CA).…”

“…The purified eCG was radiolabeled using 125-Iodine (IMS-30; Amersham, Little Chalfont, UK) according to the enzymatic procedure of Thorell and Johansson (1971). The antibody levels were expressed as the percentage of 125 I-eCG bound by 10 ml of plasma.…”

“…and used to prepare '25I EGF. A total of 19 iodinations were carried out using either iodogen (Fraker & Speck, 1978; n = 16) or lactoperoxidase (Thorell & Johnson, 1971; n = 3). The radioactive EGF thus prepared was stored in aliquots at -40°C until required, for up to 1 month.…”
