Enzyme Immunoassay: A Review

Abstract: In the last few years, the use of enzyme labels in immunoassays has been investigated. The aim of this review is to evaluate critically the role of such labels in clinical biochemistry. Special attention has been given to the problems involved in preparing enzyme labels and the ways in which such labels can be used in a variety of heterogeneous and homogeneous assay systems.

“…fluorescence quenchers, chemiluminescence precursors) have been investigated and developed in the last years (Schall and Tenoso, 1981;Oellerich, 1980). In particular, enzyme immunoassays (Borrebaeck and Mattiasson, 1979;O'Sullivan et al, 1979;Engvall and Ruoslahti, 1979; Scharpe el af., 1976; Ngo and Lenhoff, 1981;Marks, 1981) have proved to be suitable alternatives to radioimmunoassays. The various ways in which enzymes have been employed.…”

“…11α-hydroxyprogesterone-hemisuccinate BSA (molar ratio 15:1) was determined using trinitrobenzene sulfonate method (Satake, 1960). The 11α-hydroxyprogesterone-hemisuccinatehorseradish peroxidase conjugate used as labeled antigen in the competitive ELISA was prepared by the carbodiimide method (Gross and Bilk, 1968;O'Sullivan et al, 1979). 11α-hydroxyprogesteronehemisuccinate (2.46 µmol) was dissolved in 30 µl DMF, mixed with equal amounts of NHS and DCC, and the mixture was stirred for 12 h at room temperature.…”

“…An alternative method is the enzyme-linked immunosorbent assay (ELISA). After the introduction of the ELISA for the determination of rabbit IgG (2), this technique has now become used in a large and still increasing number of applications which include measurements of antigens, haptens and antibodies (3,4).…”
