Erbin inhibits TGF-β1-induced EMT in renal tubular epithelial cells through an ERK-dependent pathway

Abstract: Epithelial-to-mesenchymal transition (EMT) plays a crucial role in the progression of renal interstitial fibrosis, which finally leads to renal failure. Erbin, a member of LAP family, is recently reported to inhibit Smads and ERK pathway which are two important types of intracellular signaling involved in TGF-β1-induced EMT. However, the role of Erbin in the regulation of EMT and the underlying mechanisms remain to be fully understood. To that end, we aimed to evaluate the expression of Erbin in renal intersti… Show more

“…The MAPK pathway is a highly conserved pathway involved in diverse cellular functions, including cell proliferation, cell differentiation and apoptosis, while JNK, p38 MAPK and ERK in this pathway are involved in a Zn-sensing process [32, 33]. The results from the present study indicated that over-expression ZnT7 significantly inhibited HG-induced expression of phospho-ERK but not phospho-p38 MAPK.…”

“…These results all indicate that ACY1 functions via the ERK pathway. Furthermore, activation of ERK increases the expression of TGF-β, another important factor involved in tumor progress (13,(15)(16)(17), and TGF-β could therefore directly attenuate the immune system and promote tumor proliferation (13,17). Thus, decreased expression of TGF-β could lead to tumor inhibition.…”

“…As predicted, this revealed that silencing of ACY1 reduced the level of ERK1 expression in colorectal cancer tissue, which indicates that there is an overexpression of ERK1 along with ACY1 in colorectal cancer. In addition, since it is well known that activation of ERK leads to increased TGF-β1 expression (13,15,16), the association between ACY1 and TGF-β1 expression was measured, which revealed a positive association between the two. The results of the present study indicate that the ERK and TGF-β1 function downstream of ACY1 and facilitate the tumor-promoting effect of ACY1.…”

“…Preventing EMT-associated signaling pathways plays an important role in inhibiting cancer cell migration and invasion, as well as reducing drug resistance (12). The occurrence of the EMT is associated with complex signaling pathways, including the Wnt, PI3K, Hedgehog, transforming growth factor-β (TGF-β) and Notch pathways (16)(17)(18). However, the details of these mechanisms in HCT-8 colon cancer cells are yet to be elucidated.…”
