“…The subgingival samples were collected from the periodontitis active sites exhibiting the deepest pocket depths using three paper points that were inserted in the periodontal pockets for 60 s. The paper points were subsequently transferred into 1 mL of reduced transport fluid (RTF) and stored at −70°C, according to a modified protocol for sample collection. 17,18 The extraction of bacterial DNA from the collected samples was done using a Qiamp DNA Mini Kit (Qiagen, Hilden, Germany). Viral DNA was obtained by incubating 500 μL of samples with 10 μL of proteinase K at 55°C, and then proteinase K was inactivated by boiling at 100°C for 10 min.…”

“…The principal goal of the periodontal therapy is to restrain active inflammation during the disease and possibly provide support for the reconstruction of periodontal tissue defects [5]. Initial periodontal therapy, scaling and root planning (SRP) is not frequently resolute at repairing disease-related defects [6,7]. The periodontal wound healing after SRP usually induces the development of a long junctional epithelium, which is responsible for frequent recurrence of a periodontal pocket [8].…”

“…DNA extracts were stored at -20ºC prior to PCR analysis. Total gene copy number determination was done as described by Brajović et al [16], using Maxima™ SYBR Green/ROX qPCR Master Mix (Thermo Fisher Scientific, Waltham, MA, USA) and the following primers: Fw 5'-TCCTACGGGAGCACAGT'-3 and Rv (5'GGACTACCAGGGTATCTAATCCTGTT-3' . Real-time PCR analyses were performed on Line Gene-K Fluorescence Real-time PCR Detection System (Hangzhou Bioer Technology Co. Ltd., Hangzhou, China).…”
