“…The number of genes with significantly enhanced or reduced expression was counted for each period of follicle development (EAFs to SAFs, SAFs to LAFs, and SAFs to IVGs). In our preliminary experiments and previous reports involving the same culture methods [1, 10, 11], oocyte diameter (average 112 μm) and the ability to complete nuclear maturation (42%) were found to be similar to those in oocytes of SAFs. Accordingly, here, we compared gene expression between IVGs and SAFs.…”

“…Thus, although we could not identify a direct cause for the high responsiveness of OGCs derived from aged cows to E 2 , our results suggest that post-transcriptional modification of estradiol receptors differs between the two age groups. The responsiveness to E 2 differs among different developmental stages of follicle, and OGCs derived from advanced stage EAFs did not require much E 2 for their in vitro development [30]. In addition, we found that OGCs derived from EAFs of aged cows were at a more advanced stage compared to those of the younger counterparts.…”

“…These media were IVC medium containing 0.1 μg/mL estradiol and an IVC medium containing 10 μg/mL estradiol. Concentration of estradiol was set on the basis of the previous reports on estradiol concentration of bovine follicles [23], and that high concentration of estradiol 10 μg/mL enhanced in vitro growth of oocytes of EAFs derived from young cows and gilts [24,30]. For replicates, about 30-40 oocytes were collected from the group of young or aged cows (Table 1) and the experiment was repeated 10 times.…”

“…As estradiol reduces IGFBP3 (Huynh, Yang, & Pollak, ), use of estradiol in the medium could be reasonably expected to rescue the cultured follicles from the adverse effects. Indeed, estradiol was reported to improve survival, diameter and antrum formation in the cultured PFs in the pig (Tasaki, Iwata, Sato, Monji, & Kuwayama, ), cow (Hulshof et al., ), rat (Gore‐Langton & Daniel, ) and sheep (Kona et al., personal communication). Thus there appears to be a strong reason to counteract the inimical effects of IGFBP3 by improving the oestrogen secretion or addition of oestrogen to the cultured medium.…”
