The steroid hormone, estradiol (E 2 ), has numerous targets in the central nervous system, including the hippocampus, which plays a key role in cognition and affective behavior. This review focuses on our evidence from studies in rodents that E 2 has diverse mechanisms in the hippocampus for its functional effects E 2 has rapid, membrane-mediated effects in the hippocampus to enhance cognitive performance. Administration of E 2 to the hippocampus of rats for 10 minutes following training enhances performance in a hippocampus-mediated task. Increased cell firing in the hippocampus occurs within this short time frame. Furthermore, administration of free E 2 or an E 2 conjugate, E 2 :bovine serum albumin (BSA), to the hippocampus produces similar performance-enhancing effects in this task, suggesting that E 2 has membrane actions in the hippocampus for these effects. Further evidence that E 2 has rapid, membrane-mediated effects is that co-administration of E 2 and inhibitors of mitogen activated protein kinase (MAPK), rather than intracellular E 2 receptors (ERs) or protein synthesis, attenuate the enhancing effects of E 2 in this task. Despite these data that demonstrate E 2 can have rapid and/or membrane-mediated effects in the hippocampus, there is clear evidence to suggest that intracellular ERs, particularly the β (rather than α) isoform of ERs, may be important targets for E 2 's functional effects for hippocampal processes. Administration of ligands that are specific for ERβ, but not ERα, have enhancing effects on hippocampal processes similar to that of E 2 (which has similar affinity for ERα and ERβ). These effects are attenuated when ERβ expression is knocked down in transgenic models or with central administration of antisense oligonucleotides. Thus, there may be a convergence of E 2 's actions through rapid, membranemediated effects and intracellular ERs and in the hippocampus for these functional effects.

Anabolic-androgenic steroids (AAS) are drugs of abuse. They are taken in large quantities by athletes and others to increase performance, with negative health consequences. As a result, in 1991 testosterone and related AAS were declared controlled substances. However, the relative abuse and dependence liability of AAS have not been fully characterized. In humans, it is difficult to separate the direct psychoactive effects of AAS from reinforcement due to their systemic anabolic effects. However, using conditioned place preference and self-administration, studies in animals have demonstrated that AAS are reinforcing in a context where athletic performance is irrelevant. Furthermore, AAS share brain sites of action and neurotransmitter systems in common with other drugs of abuse. In particular, recent evidence links AAS with opioids. In humans, AAS abuse is associated with prescription opioid use. In animals, AAS overdose produces symptoms resembling opioid overdose, and AAS modify the activity of the endogenous opioid system. A search of the Google news archive in March 2007 reveals 47,500 news stories on "steroids" in 2006, a 400% increase since their debut in the archive in 2002. In 2006, "steroids" even surpassed trend-setters such as "Paris Hilton" (38,500 stories) or "spinach" (42,700 stories). Current media attention on AAS abuse has been driven by accusations of steroid use among professional athletes in baseball, track, and cycling. However, public recognition of AAS use in sports dates back nearly 20 years, when Ben Johnson tested positive for stanozolol at the Seoul Olympics in 1988 [77]. Eastern Block athletes had been using steroids since the 1950's, and the International Olympic Committee banned steroid use from Olympic competition in 1976. However, the ability to reliably detect androgen metabolites in urine samples was limited until the advent of gas chromatography-mass spectroscopy [89].
