Evaluating different extraction solvents for GC-MS based metabolomic analysis of the fecal metabolome of adult and baby giant pandas

Abstract: The gut microbiome plays a fundamental role in host health and the fecal metabolome can be analysed to assess microbial activity and can be used as an intermediate phenotype monitoring the host-microbiome relationship. However, there is no established extraction protocol to study the fecal metabolome of giant pandas. The aim of this research is to optimize extraction of the fecal metabolome from adult and baby pandas for high throughput metabolomics analysis using gas chromatography-mass spectrometry (GC-MS). … Show more

“…Fecal water was extracted by performing a successive two-solvent (ethanol and Diethyl ether)-based metabolite extraction protocol. It has been reported in a recent study that extraction efficiency was high for ethanol or methanol and isopropanol protocols [15,16]. As methanol is more carcinogenic than ethanol, we chose to use ethanol as an extraction solvent.…”

“…Although knowledge of the stability of the analytes in DBS is of crucial importance for biomarker discovery, data available on the assessment of human DBS stability for metabolomics analysis are still scarce. Existing publications on untargeted metabolomics mention several protocols intended to improve the metabolome coverage [37,38]. Yet, few targeted large cohorts of patients and, to our knowledge, none focused on disease-specific metabolites.…”

“…This protocol was in accordance with the previous study ( Yang Y. et al, 2019 ), with minor modifications. First, fecal samples ( n = 10/8, control/CSDS) were dehydrated in a SpeedVac (Thermo SPD111V) for 3 h and weighed into 2 mL screw-cap tubes, for extraction.…”
