Evaluation of a commercial test for antibodies to the chlamydial lipopolysaccharide (MedacTM) for serodiagnosis of acute infections by Chlamydia pneumoniae (TWAR) and Chlamydia psittaci

Abstract: A commercial test (rELISA) based on a recombinant chlamydial lipopolysaccharide (LPS) antigen has been evaluated for the diagnosis of acute infections caused by Chlamydia pneumoniae (TWAR) and Chlamydia psittaci. This test and a microimmunofluorescence test (MIF) were compared in 160 patients with community-acquired pneumonia. Seventeen of nineteen cases with significant titre changes detected by rELISA were confirmed by MIF. The two remaining cases not confirmed by MIF were considered false-positive reactions… Show more

“…These rough mutants rarely occur in nature, and the rELISA is therefore considered genus specific (9). Nevertheless, cross-reactivity with M. pneumoniae and parvovirus has been reported (39). Of the four samples with false-positive results by the rELISA, none was positive for M. pneumoniae and two were positive for parvovirus; one of the latter two samples was also positive for C. trachomatis and C. psittaci (Tables 1 and 2).…”

“…Thus, in total, 34 samples (43%) were positive. The cross-reactivity of the rELISA with parvovirus and M. pneumoniae might account for false-positive results (39). Therefore, the four samples negative for C. pneumoniae-specific IgG antibodies but positive by rELISA were tested for antiparvovirus-specific and anti-M. pneumoniae-specific IgG.…”

“…Serological testing was performed on two consecutive sera drawn at least 2 weeks apart (Chlamydia IgG/A/M rELISA; Medac Diagnostika). A psittacosis case was considered serologically proven by a threefold rise in Chlamydia spp.-specific IgG, a twofold or greater change in the specific IgM, or a twofold increase in the specific IgG titre, in combination with a twofold increase in the specific IgA antibody titre (Persson & Haidl, 2000;Verkooyen et al, 1997Verkooyen et al, , 1998. The CFT was performed with a commercially available Chlamydia group antigen (Virion) on all recombinant ELISA (rELISA)-positive sera.…”
