Evaluation of automated molecular tests for the detection of SARS‐CoV‐2 in pooled nasopharyngeal and saliva specimens

Abstract: Background Pooling of samples for SARS‐CoV‐2 testing in low‐prevalence settings has been used as an effective strategy to expand testing capacity and mitigate challenges with the shortage of supplies. We evaluated two automated molecular test systems for the detection of SARS‐CoV‐2 RNA in pooled specimens. Methods Pooled nasopharyngeal and saliva specimens were tested by Qiagen QIAstat‐Dx Respiratory SARS‐CoV‐2 Panel (QIAstat) or Cepheid Xpert Xpress SARS‐CoV‐2 (Xpert), and the results were compared to that of… Show more

“…The relative fold change data were calculated using the comparative Ct method, often known as 2 ΔΔCt . Following the biological averaging strategy used in other research with real-time PCR, microarray, and RNA-Seq analysis, three pooled biological replicates and three technical replicates were used in qPCR experiments to upsurge statistical reliability [ 57 , 58 , 59 ].…”

“…Automation can also contribute to accurate testing of pooling procedures by providing accurate mixing, sample tracking, and avoidance of cross-contamination. In previous reports, the pooling and testing stages were usually performed manually (or methods not specified), although some studies utilized automated extraction devices or fully automated platforms for the testing of pooled samples ( 4 , 16 19 ). Barak et al utilized a liquid handler for automated media pooling but its performance was not validated by comparison with individual testing ( 7 ).…”

“…Representative POC versions of the Abbott ID NOW COVID-19, Xpert Xpress SARS-CoV-2, and AcculaSARS-CoV-2 Test have been approved for emergency use 12 . However, the existing detection modules have low throughput, which restricts their use in large-scale testing in a short interval 13,14 . Moreover, the high mutation rate and the emergence of new SARS-CoV-2 variants necessitate continuous updates in PCR diagnostic tests that should be able to detect multiple SARS-CoV-2 targets and not be in uenced by genetic variations.…”
