Evaluation of different recording parameters to establish a standard for flash electroretinography in rodents

Abstract: Different electrodes and stimulus protocols commonly used for electroretinography in rodent eyes were compared for convenience of use, degree of damage to corneal epithelium, and for magnitude of amplitude, reproducibility, left versus right eye accuracy, and reliability of recorded parameters of the flash electroretinogram (ERG). Adult C57BL/6 pigmented mice and albino Wistar rats were used to determine scotopic ERGs in response to Ganzfeld or strobe-light stimulation and light-adapted (photopic) ERGs recorde… Show more

“…If the cornea becomes dehydrated, corneal scarring and cataract formation may occur 24 . Cataract formation is more common in mice 25 , and various methods of maintaining corneal hydration have been employed in mouse ERG recordings, including a constant flow of aqueous fluid or custom-made contact style electrodes that trap hydration at the corneal surface 12 .…”

“…Bayer and collaborators [8] demonstrated that the contact of a metal wire loop electrode (stainless steel wire loop) injures the rodent cornea, whereas the cotton contact is less aggressive (using a cotton-silver electrode). Our cotton electrode enables repeated recordings over time.…”

“…The waveform obtained depends upon the recording conditions, the body temperature [7] , the light stimulator [8] and the anesthesia used [9] . The response obtained is also highly dependent on the recording electrode used.…”
