Influence of Growing Region on Targeted Anthocyanin Profile, Color, Phenolics, and Antioxidant Activity of Grape Juice cv. Manjari Medika (MM) were examined. The prevailing environmental conditions affect chemical composition or metabolites of grapes. The MM grape berries were harvested from three environmentally different locations in India i.e. Pune (MMJP), Vijayapura (MMJV), and Nashik (MMJN). MMJV, grown in region with high average temperatures and low rainfall showed higher levels of (393.65 ± 8.18 g/L) reducing sugar with lower levels of color intensity (49.69±0.55%), juice recovery (68.70±0.54%), and acidity (6.98±0.04 g/L) followed by flavonoids (0.023 g/L) than the regions with relatively low temperatures and high rainfall ((MMJP) and (MMJN)). In UHPLC-Oribitrap MS analysis, a total of 11 anthocyanins, which includes delphinidin-3-galactoside (Dp3Ga), cyanidin (Cy), peonidin-glucoside-4-Vinylphenol (Pn3Glu4VP), petunidin-3-glucoside (Pt3Glu), malvidin-3-glucoside (Mv3Glu), pelargonidin (Pg), petunidin-3-acetyl glucoside (Pt3ace), peonidin-3-acetyl glucoside (Pn3ace), malvidin-3-acetyl glucoside (Mv3ace), delphinidin (Dp) and petunidin (Pt) were detected in grape juice samples. MMJV. It is interesting to note that among the 11 anthocyanins detected, two compounds, petunidin-3-acetyl glucoside (Pt3ace) and peonidin-3-acetyl glucoside (Pn3ace), have been reported only in specific grape juice samples, i.e., Pt3ace in MMJV and Pn3ace in MMJN. Discriminatory compounds among the regions were petunidin-3-acetyl glucoside (Pt3ace), peonidin-3-acetyl glucoside (Pn3ace), and cyanidin (Cy) according to principal component analysis (PCA). UHPLC-MS results showed that the total anthocyanin content of the fresh juice in the cultivar tended to increase with altitude from sea level.

The by-products of the food industry can be used as nutrient source to improve the nutritional characteristics of other food products. This study aimed to investigate the chemical composition, antioxidant activity and phenolic compound content of organic grape peel flour in cookies using response surface methodology. Composition analysis using different assays (DPPH, ABTS, FRAP, ORAC) revealed that this flour was low in calories (180.22 AE 0.25 kcal%), with low dietary fibre content (50.10 AE 0.01 g%) and high antioxidant capacity. The total phenolic compound analysis showed high values of phenolic compounds (1063.58 AE 8.09 mg%), consisting of resveratrol (54.27 AE 0.79 mg%), epicatechin (43.94 AE 0.90 mg%) and chlorogenic acid (35.70 AE 1.47 mg%). A 2 3 factorial experimental design was formulated to optimise the ingredients for cookie production. The resulting cookies presented high acceptance and good sensory properties, confirming that organic grape flour did not interfere with product acceptability. Thus, cookies with an acceptable quality and improved nutrition can be prepared using organic grape peel flour.

Objective Grape Seed Extract is a natural source of various polyphenols, which have been shown to possess potent antioxidant and free radical-scavenging activities. The earlier studies have reported that grape seed extract exhibits broad-spectrum pharmacological activities. Therefore, studying the hepatoprotective effects and elucidation of mechanisms of action of the Indian Variety, Manjari Medika grape seed extract (GSE), may give an insight into therapeutic benefits. Methotrexate (MTX) is the first-line pharmacological therapy for different rheumatic diseases. The major adverse events such as hepatotoxicity are evident even in the low doses used for the treatment. The present study investigated the role of MTX on hepatic damage in murine liver and the plausible protective effects of the Indian grape variety, Manjari Medika grape seed extract, in ameliorating it. Methods and Results To assess the hepatological modulation, mice were divided into eight groups to investigate the ameliorative potential of this GSE (75 and 125 mg/kg) and correlate the experimental findings. The active components of the extract were assessed through UPLC-(ESI)-QToF-MS analysis. On the other hand, various biochemical and immunological indices were carried out to correlate the experimental data. The result demonstrated that the prophylactic administration of GSE reduced MTX-induced hepatic toxicity indices, which subsequently restored the hepatic morphological architecture. Moreover, the application of GSE in a dual dosage (75 and 125 mg/kg) suppressed MTX-induced reactive oxygen species generation, followed by lipid peroxidation and cellular nitrite formation. MTX-induced inflammasome activation through the redox-assisted cascade of TLR4/NF-κB signaling was further reduced by applying the GSE. The results showed that the activation of cytoprotective transcription factor Nrf2 enhanced the level of endogenous antioxidants. Furthermore, through the regulation of TLR4/NF-κB and Nrf2/HO-1 axis, this extract could reduce the MTX-mediated hepatic damage. Conclusion Our findings suggest that Manjari Medika seed extract could be used as a therapeutic agent to relieve the side effects of MTX and other hepatic disorders.
