Newcastle disease (ND) is an infectious, highly contagious and lethal disease of avian species. It is considered that ducks are natural reservoir or carrier for Newcastle disease virus (NDV) and are resistant against different strains of NDV. Current study was designed to evaluate the pathogenesis of Newcastle disease in domestic ducks through histopathology, immunohistochemistry (IHC) and serum biochemical changes. For this purpose, eighty ducks were reared for 42 days and divided in two groups A and B. Ducks in group A were challenged with (NDV) at rate of 0.1 ml of ELD50 (virus titer 107.32/100µl) on second week of age, whereas Group B was control negative. Splenomegaly, atrophy of thymus and necrotic lesion in kidney were observed on 9th day of post infection. Hepatic degeneration and mononuclear cell infiltration were noticed in proventriculus and intestine in challenged ducks. Viral antigen detected in lungs, intestine, proventriculus and lymphoid organs of infected ducks through IHC. Albumin and total protein values were significantly low in infected groups A as compared to control group B. ALT, AST, and ALP values were significantly high in infected group A. On 5th and 7th day of post infection oropharyngeal swabs were negative for NDV and cloacal swabs were positive for NDV through Reverse transcriptase polymerase chain reaction. It is concluded that ducks are susceptible to NDV and virulent strain of NDV caused disease in ducks.

Penelitian ini bertujuan untuk mengetahui perubahan patologi anatomi dan histopatologi organ timus, bursa Fabricius, dan limpa itik bali yang diinfeksi virus newcastle disease atau Avian orthoavulavirus 1 isolat Tabanan-1/ARP/2017. Penelitian ini menggunakan 15 ekor itik bali jantan berumur satu minggu. Setelah periode aklimatisasi selama dua minggu dan pengukuran titer antibodi ND dalam serum itik menunjukkan hasil di bawah angka 23 HI unit, selanjutnya sejumlah 10 ekor itik diinfeksikan dengan AOAV-1 isolat Tabanan-1/ARP/2017 dengan dosis 0,5ml/27 HA unit secara intraokular (P1) dan 5 ekor itik lainnya diberikan 0,5 ml larutan phosphate buffered saline melalui rute yang sama sebagai kontrol (P0). Nekropsi kemudian dilakukan pada itik yang mati dan dilakukan pemeriksaan patologi anatomi terhadap organ pertahanannya. Selanjutnya organ tersebut diproses dan dipulas dengan pewarnaan Hematoksilin-Eosin, lalu dilakukan pemeriksaan histopatologi. Hasil penelitian menunjukkan bahwa infeksi AOAV-1 isolat Tabanan-1/ARP/2017 pada itik bali dapat menyebabkan perubahan patologi anatomi berupa lesi foki nekrotik pada limpa dan atrofi pada timus. Sementara pada gambaran histopatologi menimbulkan deplesi sel limfoid berderajat parah pada organ timus dan bursa Fabricius, serta deplesi sel limfoid berderajat ringan pada organ limpa.

Although waterfowl are less susceptible to Newcastle disease (ND) virus (NDV) infection compared with chickens and turkeys, lethal ND in waterfowl has been sporadically reported. Factors underlying the high pathogenicity of certain NDV strains in waterfowl remain unclear. In ducks, the NDV 9a5b isolate shows low pathogenicity while the d5a20b isolate shows high pathogenicity. This study aimed to identify the definitive lesions that led to the lethal virulence of d5a20b by comparing the histopathology of 9a5b- or d5a20b-inoculated ducks in order to elucidate lesions related to the enhanced pathogenicity of certain NDV strains in ducks. Herein, 7-day-old ducks were intranasally inoculated with either 9a5b or d5a20b NDV strains. The neurological signs were more severe in the d5a20b-inoculated group than in the 9a5b-inoculated group. Ducks in the d5a20b-inoculated group exhibited more severe lymphoid depletion in immune organs than those in the 9a5b-inoculated group, which may have caused an immunosuppressive state in the d5a20b-inoculated ducks. Ducks in the d5a20b-inoculated group had more severe nonsuppurative encephalitis with increased NDV nucleoprotein than those in the 9a5b-inoculated group. Additionally, pancreatic necrosis, with intralesional NDV nucleoprotein, was more severe in the d5a20b-inoculated group than in the 9a5b-inoculated group. Our results showed that the immune organs, brain, and pancreas were significant targets of the NDV d5a20b infection in ducks.
