Evaluation of miRNA Detection Methods for the Analytical Characteristic Necessary for Clinical Utilization

Abstract: miRNAs are promising biomarkers but methods for their measurement are not clear. We therefore examined three miRNA detection technologies and considered the analytical characteristics essential for clinical utilization. TaqMan assays, SplintR-qPCR and miREIA were compared for their absolute quantification bias, conformity and robustness. Absolute concentrations of miR-142-5p, miR-23a-3p and miR-93-5p were measured with all three methods using 30 samples. Robustness was evaluated by measurement of miR-21-5p in … Show more

“…Previous nanopore-based studies reported sensitivities primarily in the nM range, with a few at pM range, which are insufficient for direct detection in biofluids and therefore requires multiple pre-enrichment or concentration steps 29 , 31 , 49 , 50 . The fM sensitivity reported here represents a major improvement over current single-molecule 21 , 51 or general electrochemical and fluorescence-based 35 techniques and is comparable to amplification-based RT-qPCR 52 . Due to the single-molecule resolution, as little as 5 zeptomol (ca.…”

“…Previous data was obtained with RT-qPCR while ours was obtained with miREIA. The miREIA technique is fast, specific and has great correlation with RT-qPCR [ 21 ]. Importantly, it enables easy application of microRNA biomarkers in clinical laboratories that do not have a PCR machine.…”

“…However, apart from the sensitivity and specificity of the detection, the necessary microarray equipment is fairly expensive as well as the required personnel expertise, and therefore, microarray platforms are rather suitable for miRNA expression surveys concerning a certain disease (Krepelkova et al, 2019;Ouyang et al, 2019).…”
