Evaluation of Mycobacterium avium subsp. paratuberculosis isocitrate lyase (IcL) and ABC transporter (BacA) knockout mutants as vaccine candidates

Abstract: There has been little success in controlling Johne’s disease, caused by Mycobacterium avium subsp. paratuberculosis, due to suboptimal diagnostics and the ineffectiveness of available vaccines. By knocking out BacA and IcL, genes required for MAP survival in dairy calves, two live-attenuated vaccine candidates were created. This study evaluated the host-specific attenuation of MAP IcL and BacA mutants in mouse and calf models, as well as the elicited immune responses. Deletion mutants were generated in MAP str… Show more

“…This assay showed that the macrophages from animals vaccinated with prototype vaccines, cultured alone or in a co-culture with unprimed PBMCs, showed an increased MAP-killing ability. This is in agreement with a more effective clearance of MAP infection from calf tissues in vivo [98].…”

“…These kinds of models can give information about the crosstalk between the cell types evaluated and also about the hostpathogen interactions. Therefore, most ex vivo assays described in the literature have focused on macrophages, either alone or in co-culture with other cell types, including lymphocytes [96,97], PBMCs [98], neutrophils [94], or even epithelial cell lines [99,100]. The objective of each of these co-culture models varies depending on the cell type and on the origin of the cells (vaccinated, healthy, or MAP-infected animals).…”

“…Previous studies indicated that the C57BL/6 mouse model does not reflect JD in ruminants [ 42 , 43 ]. Moreover, this mouse model showed a decrease in MAP CFUs over time in the liver compared with the less immunocompetent BALB/c mice, which is an indication that C57BL/6 mice are somewhat resistant to infection [ 20 ].…”
