Evaluation of simple and complex sensorimotor behaviours in rats with a partial lesion of the dopaminergic nigrostriatal system

Abstract: We have examined the behavioural consequences of a partial unilateral dopaminergic denervation of the rat striatum. This partial lesion was obtained by an intrastriatal 6-hydroxy-dopamine injection (6-OHDA, 20 or 10 microgram divided between two injection sites) and was compared with a unilateral complete lesion resulting from an injection of 6-OHDA (2 x 6 microgram) into the medial forebrain bundle. Quantification of striatal dopamine (DA) and its metabolites, and the immunohistochemical evaluation of the nig… Show more

“…The reduction in the number of pellets eaten on the side contralateral to the lesion in the rats with a 6-OHDA lesion is in line with a massive body of studies utilising this task as a measure of skilled reaching after striatal DA depletion (Barneoud et al, 2000;Jeyasingham et al, 2001;Klein et al, 2007;Samoudi et al, 2012;Whishaw et al, 1997). There was however no difference between the rats with a pure DA depletion and those with the additional depletion of 5HT and NA, which readily matches the findings from the movement time in the lateralised CRT task.…”

“…Furthermore, reductions of the number of TH positive varicosity profiles in striatal subfields, after the injection of 8 µg of 6-OHDA, were described in our previous paper using a stereologic method for quantification (Rodrigues et al, 2001). These findings are in agreement with previous reports that have demonstrated decreases in the striatal dopamine levels and in the striatal TH immunoreactivity after a local injection of 6-OHDA (Lee et al, 1996;Kirik, Rosenblad, & Björklund, 1998;Chang, Wachtel, Young, & Kang, 1999;Barnéoud, Descombris, Aubin, & Abrous, 2000;Rosenblad et al, 2000). Our findings show that a decreased number of dopamine neurons in the ipsilateral VTA, after a single, unilateral injection of 6-OHDA in the striatum, are in full agreement with previous reports (Cadet & Zhu, 1992;Przedborski et al, 1995;Lee et al, 1996;Kirik et al, 1998).…”

“…Motor impairments typically associated to clinical symptoms in human PD patients have been observed in rodents treated with MPTP or 6-OHDA. A complete lesion of SNc by 6-OHDA injection impaired dramatically (by 80%) the stepping of the contralateral paw (Barnéoud et al, 2000). Animals with more than 95% bilateral SNc lesion demonstrated bradykinesia symptoms (Sakai and Gash, 1994) and ≈75% DA cells loss produced forelimb akinesia manifested in stepping deficit (Tseng et al, 2005).…”
