Eelgrass meadows have attracted a lot of attention not only for their ability to maintain marine ecosystems as feeding grounds for marine organisms, but also for their potential to store atmos-pheric and dissolved CO2 as blue carbon. This study comprehensively evaluated the bacterial and chemical composition data obtained from eelgrass sediments of different scales along the coast of Japan to investigate the sediment effect on the acclimatization of eelgrass. Regardless eelgrass habitat, approximately 1% Anaerolineales, Babeliales, Cytophagales, and Phycisphaerales were present in the bottom sediment. Sulfate-reducing bacteria (SRB) were present at 3.69 % in eelgrass sediment compared to 1.70 % in bare sediment. Sulfur-oxidizing bacteria (SOB) were present at 2.81% and 1.10% in the eelgrass and bare sediment, respectively. Bacterial composition analysis and linear discriminant analysis revealed that sulfur-oxidizing bacteria detoxified H2S in the eelgrass meadows and that the larger-scale eelgrass meadows had a higher diversity of sul-fur-oxidizing bacteria. Our result indicated that one of the following systems detoxifies the H2S in eelgrass meadows microbial oxidation by sulfur-oxidizing bacteria, oxidation by O2 perme-ated by the physical diffusion of benthos. This study indicated the issues of maintaining sustain-able conservation of the eelgrass bed.

Eelgrass meadows are vital not only for sustaining marine biodiversity as marine ecosystems but also for carbon fixation as blue carbon. However, the national decline in eelgrass meadows in Japan, which was initiated by economic growth in the 1980s, prompted the implementation of conservation measures. This study explored the fundamental techniques of the sediment environment to enhance Zostera marina seed germination rates. We investigated the microbial origin of Zostera marina seeds under anaerobic conditions and assessed their germination efficiency. Anaerobic sediments in the eelgrass meadows were found to increase eelgrass germination rate by 6% more than aerobic sediments. And interestingly, the sulfate-reducing bacteria Desulfovibrionaceae and Sulfurospirillaceae were detected mainly on eelgrass seeds. Our result clearly indicated that the sulfate-reducing bacteria around Zostera marina seeds create anaerobic conditions that promote germination through their sulfate-reducing function. Therefore, when developing new eelgrass meadows, it can be expected to improve germination rates by preparing the sediment environment to facilitate the increase of SRB coexisting with seeds. However, the difference in bottom sediment conditions varies by region, so it is necessary to be careful when adding or removing something. In addition, germination and growth of eelgrass must be considered separately, and factors that promote growth are not described in this work. The result of this study was an important finding for the implementation of Blue Carbon activities to improve sediment quality prior to the creation of eelgrass meadows and can also support eelgrass meadows maintenance after establishment.

The success of a seagrass transplantation effort depends on identifying locations with optimal sediment characteristics for seagrass growth and survival. This study analyzed sediment characteristics of seagrass bed sediment and adjacent unvegetated sediment on Lae-Lae Island and Lae-Lae sandbar (Makassar, South Sulawesi) to determine the suitable sites for seagrass transplantation. Seagrass bed sediments and adjacent unvegetated sediments were collected from four locations to measure sediment particle size, redox potential, organic matter, water content, nitrate, and phosphate concentrations. Analysis of similarities (ANOSIM) was performed using PAST (Paleontological Statistics) software to determine a suitable site for seagrass planting (transplantation). The results show that surface sediments in the Lae-Lae island are composed of medium sand and coarse sand with grain sizes varying from 0.290 to 0.768 mm, whereas in the Lae-Lae sandbar, the sediment is composed of medium sands (0.371 – 0.460 mm). Redox potential (Eh) varied from -34.1 to -65.7mV, water contents were 1.3 to 1.8%, organic contents were 19.85 to 38.53%, nitrate content varied from 0.60 to 3.47, and phosphate content varied from 7.42 to 14.19 ppm. The percentage of mud (clay and silt) and organic matter were slightly higher in seagrass bed sediments compared to unvegetated area sediments. No differences in nutrient contents (nitrate and phosphate) between seagrass sediments and unvegetated were observed in this study. ANOSIM results show no difference in the sediment characteristics between a seagrass bed and unvegetated sand at the north and south sides of Lae-Lae Island and Lae-Lae sandbar, which means that these three sites are suitable for seagrass transplantation.Keywords: sediment, seagrass, transplantation, site selection, ANOSIM, Lae-Lae
