Evidence for Increased Lipid Peroxidative Damage and Loss of Superoxide Dismutase Activity as a Mode of Sublethal Cryodamage to Human Sperm During Cryopreservation

JUAN G. ALVAREZ,
BAYARD T. STOREY

Abstract: Cryopreservation of human sperm, now generally required in donor insemination programs, adversely affects the sperm in terms of standard sperm evaluation parameters and fertilizing ability. The freeze‐thaw process appears to produce sublethal damage that appears only after a delay. The authors hypothesized that cryopreservation enhanced peroxidation of sperm membrane lipids, based on previous studies of sperm lipid peroxidation, which showed that the effects of peroxidative damage became evident only after a d… Show more

“…Alguns fatores decorrentes da refrigeração como choque térmico e estresse osmótico podem causar injúrias a célula espermática que leva a perda da fertilidade e da capacidade do espermatozoide fecundar (Mazur, 1984;Storey, 1992;Amirant, et el., 2004;Li et al, 2005).…”

“…Antioxidant enzyme activities of mammalian spermatozoa comprise those provided by superoxide dismutase (SOD) and GPX. SOD has been shown to play a key role in the intracellular scavenging of superoxide anions [ 145 ]. The superoxide anion generated by the inner mitochondrial membrane is dis-mutated to hydrogen peroxide by the Mn-SOD present in the mitochondrial matrix and by the Cu/Zn-SOD present in the cytosol.…”
