Evoked potentials, reaction time and cognitive performance in on and off phases of Parkinson's disease.

Abstract: SUMMARY Seven patients with Parkinson's disease and severe motor fluctuations were studied with event-related potentials, reaction and movement times and neuropsychological tests while in "off' and "on" phases. While there was a significant decrement in the P300 latency of the event-related potentials and in the duration ofmovement time in the "on" phase, no significant differences between on and off phases were observed in reaction time and neuropsychological tasks.

“…In most of the studies on levodopa effects, nevermedicated PD patients or patients who underwent a drug washout period (12 h -14 days) were tested on an oddball procedure, first off and then on medication. We identified nine studies employing such a study design with five of them demonstrating a reduction of P3b latency following levodopa administration (Lukhanina et al, 2009;Oishi et al, 1996;Sohn et al, 1998;Stanzione et al, 1991;Starkstein et al, 1989). However, P3b latency was not significantly decreased by levodopa in the studies by Chia et al (1995), Kobayashi et al (2004), and Mathis et al (2014) and even increased under levodopa in the study by Prasher and Findley (1991).…”

“…To summarize the findings, after an administration of L-dopa, P3 latency was shortened parallel to RT only in patients with longstanding PD who, eventually, were suffering from motor fluctuations (Starkstein et al, 1989;Stanzione et al, 1991). On the contrary, in younger patients with early PD, the P3 wave was delayed after L-dopa administration (Prasher and Findley, 1991).…”

“…Starkstein et al (1989) patients suffering from severe motor fluctuations, all of them being treated by L-dopa. The ERP recording was done twice in the same day-when the patients were "off' after an overnight withdrawal of the drug, and when optimal motor status ("on") was reached after reintroduction of the treatment.…”

“…Так, S. Starkstein и соавт. [23], а также D. Prasher и L. Findley [24] установили, что у пациентов после приема леводопы латентность пика Р300 существен-но уменьшается. Аналогичные данные получены и в исследовании P. Stanzione и соавт.…”
