Exogenous Antioxidants Improve the Accumulation of Saturated and Polyunsaturated Fatty Acids in Schizochytrium sp. PKU#Mn4

Abstract: Species of Schizochytrium are well known for their remarkable ability to produce lipids intracellularly. However, during their lipid accumulation, reactive oxygen species (ROS) are generated inevitably as byproducts, which if in excess results in lipid peroxidation. To alleviate such ROS-induced damage, seven different natural antioxidants (ascorbic acid, α-tocopherol, tea extract, melatonin, mannitol, sesamol, and butylated hydroxytoluene) were evaluated for their effects on the lipid accumulation in Schizoch… Show more

“…Positive effects of ascorbic acid on the growth and DHA production by Schizochytrium sp. have been reported [ 17 , 18 ]. The results were used to identify strategies for enhancing the production of phospholipids and carotenoids in repeated-batch cultures of RT2316-16.…”

“…Several strategies have been developed for alleviate ROS for increasing lipid content. For example, addition of antioxidants, such as ascorbic acid or mannitol for enhancing fatty acid production ( Ren et al, 2017 ; Han et al, 2021 ); overexpression of oxidative stress defense genes to promote cell growth and lipid yield ( Zhang et al, 2018 , 2021 ). In our study, the ROS level was determined in Schizochytrium after adding 6.0 g/L of propanol or 40.0 g/L of 1,3-PD.…”

“…showed enhanced saturated and polyunsaturated fatty acids with a yield of 103.7% and 49.6% increment, respectively. Transcriptome changes investigated by RNA-sequencing revealed differential expression of lipid biosynthetic genes, in addition to transcripts encoding enzymes that catalyze the biosynthesis of steroids and carotenoids [ 31 ]. The addition of exogenous antioxidants resulted in the up-regulation of catalase, a key antioxidant enzyme that mediates intracellular ROS scavenging.…”

“…It is possible that ascorbic acid can up-regulate the expression of genes involved in fatty acid synthesis, leading to increased levels of unsaturated fatty acids in the membrane. An increase in the accumulation of saturated and unsaturated fatty acids in algae exposed to antioxidants was reported [ 31 ]. Analysis of differential genes involved in the process of lipid biosynthesis revealed up-regulation of two transcripts of the ketoacyl-reductase gene involved in the biosynthesis of fatty acids, two transcripts encoding diacylglycerol pyrophosphate phosphatase 1, and one transcript encoding glycerol-3-phosphate O-acyltransferase.…”
