Exosomes in atherosclerosis: performers, bystanders, biomarkers, and therapeutic targets

Abstract: Exosomes are nanosized lipid vesicles originating from the endosomal system that carry many macromolecules from their parental cells and play important roles in intercellular communication. The functions and underlying mechanisms of exosomes in atherosclerosis have recently been intensively studied. In this review, we briefly introduce exosome biology and then focus on advances in the roles of exosomes in atherosclerosis, specifically exosomal changes associated with atherosclerosis, their cellular origins and… Show more

“…Zheng et al found that vascular smooth muscle cellderived exosomes mediate the transfer of Krüppel-like factor 5induced miR-155 from smooth muscle cells to endothelial cells, damaging the tight junction of endothelial cells and the integrity of barriers leading to increased endothelial cell permeability and accelerating atherosclerosis (76). Macrophage-derived exosome miR-21-3p, which has been treated with nicotine, the principal component of cigarette smoke, may accelerate the development of atherosclerosis by increasing the migration and proliferation of vascular smooth muscle cells through its target protein (77). Exosomes mediate the occurrence and development of CVDs via upstream or downstream signaling pathways in vivo, and the relevant mechanisms are being elucidated (78)(79)(80).…”

“…miRNA-133a, miRNA-143/145, miRNA-150, miRNA-155, miRNA-214, miRNA-223, and miRNA320b in exosomes are considered as markers of arthrosclerosis (AS) [ 131 , 132 ]. Several proteins are suggested as biomarkers of AS, including integrins derived from exosomes secreted via macrophage foam cell [ 133 ], PSMA6 (prostate-specific membrane antigen), PSMA7, and annexin A2 in blood exosomes originated from innate immune system cells [ 21 , 134 ] and VCAM-1 (vascular cell adhesion molecule 1) and eNOS (endothelial constitutive nitric oxide synthase) identified in plasma endothelial cell-derived exosome [ 131 , 135 ].…”

“…Ox-LDL induces monocytes/macrophages inflammatory response emerge as a key event in atherosclerosis. EVs released from macrophages stimulate pro-inflammatory responses by transporting miRNAs and lncRNAs into recipient cells (88). For example, M1-like-type macrophages secrete numerous pro-inflammatory EVs, with anti-angiogenic properties (45).…”

“…As for diagnosis, miRNAs and lncRNAs in EVs have important advantages of serving as non-invasive biomarkers due to EVs are easily accessible and highly stabile in body fluids, such as blood, saliva, and urine (88). In addition, miRNAs and lncRNAs encapsulated in EVs exhibit better application compared with circulating miRNAs and lncRNAs, due to the high sensitivity and specificity of EVs (128).…”
