Experimental infection with Chlamydia pneumoniae in nonhuman primates

Abstract: To serially examine the immunopathogenesis and histopathology of infection with Chlamydia pneumoniae, we inoculated two cynomolgus monkeys in the conjunctival sac, nose, and nasopharynx with C. pneumoniae TWAR. After inoculation, C. pneumoniae was isolated from the inoculation sites and the rectums of both monkeys for a period of 5 weeks. After a second inoculation, C. pneumoniae was recovered from the inoculation sites and the rectums of both monkeys for 20 weeks. A third inoculation with C. pneumoniae caused… Show more

“…Two specimens were obtained from each monkey prior to inoculation with C. pneumoniae; and 12 specimens were obtained at weeks 1, 4, and 18 post-inoculation (p.i.). A reinoculation occurred at week 15 (17). For these clinical specimens, PCR products were detected by PCR-enzyme immunoassay (3).…”

“…In the nonhuman primate models (baboons and cynomolgus monkeys), mild or asymptomatic chronic respiratory tract infection (mild interstitial pneumonia) developed after intranasal inoculation (1,8). Recently, Moazed et al (18) has described C. pneumoniae infections of the rabbit.…”

“…Cell lines. RAW 264.7 cells (termed RAW cells), a murine macrophage cell line (ATCC CRL-24), and HEp-2 cells were maintained in Dulbecco medium supplemented with 10% fetal calf serum, glutamine, and antibiotics (19). U937 cells, a nonadherent human macrophage line, and McCoy cells were maintained in RPMI 1640 supplemented as described above.…”

“…After centrifugation for 1 h at 37ĐŠC at 800 Ď« g, the inoculum was removed and growth medium containing 1 g of cyclohexamide per ml was added. For McCoy cells and HEp-2 cells, growth medium was supplemented RPMI 1640 or Iscove's Dulbecco medium (19). For smooth muscle cells, smooth muscle growth medium (Clonetics) was used.…”
