Explaining the Serological Characteristics of Streptococcus suis Serotypes 1 and 1/2 from Their Capsular Polysaccharide Structure and Biosynthesis

Abstract: The capsular polysaccharide (CPS) is a major virulence factor in many encapsulated pathogens, as it is the case for Streptococcus suis, an important swine pathogen and emerging zoonotic agent. Moreover, the CPS is the antigen at the origin of S. suis classification into serotypes. Hence, analyses of the CPS structure are an essential step to dissect its role in virulence and the serological relations between important serotypes. Here, the CPSs of serotypes 1 and 1/2 were purified and characterized for the firs… Show more

“…However, this property was not observed herein using a ST25 strain: as such, it would be interesting to confirm using other cell types. It is important to note that composition and structure are responsible for serotype determination [ 42 , 46 ], and that all serotype 2 strains have the same CPS antigenically, which indicates a highly similar structure and composition. In addition, the cps locus is identical in all serotype 2 strains sequenced to date, regardless of sequence type [ 47 ].…”

“…Depending on the sensitivity of the specific CPS to hydrolysis, two methods of CPS extraction were used. For serotypes 2, 1, 1/2, and 3, the autoclave method was used (23,28,30).…”

“…Goyette-Desjardins et al [51] characterized the protective activity of monoclonal antibodies (mAbs) directed against S. suis serotype 2 CPS. All mAbs targeted the sialylated chain of the CPS, but showed different cross-reactivity against S. suis serotypes 1, 1/2, and 14, whose CPS structures are similar to that of serotype 2 [52][53][54]. Only the immunoglobulin (Ig)G1 mAb was shown to be serotype 2-specific.…”
