Expression analysis of BCL2L12, a new member of apoptosis-related genes, in colon cancer

Abstract: Apoptosis is an active process regulated by a variety of genes. Recently, the molecular cloning, physical mapping and expression analysis of a novel gene of the Bcl-2 family, BCL2L12, was reported. Expression analysis of the BCL2L12 gene in breast cancer confirmed an association of BCL2L12 with favorable prognosis of patients. In the present study, the expression of the BCL2L12 gene was analyzed in colon cancer by RT-PCR. Two transcripts, BCL2L12 and BCL2L12-A, were overexpressed in the cancer tissues as compa… Show more

“…27 A recent study also showed that NPC patients with BCL2L12 mRNA-positive tumors have remarkably shorter disease-free survival than those with BCL2L12 mRNAnegative tumors. 28 More recently, Papageorgious et al (2011) 29 also reported an unfavorable prognostic value of BCL2L12 in chronic lymphocytic leukemia in terms of overall survival, which implies an anti-apoptotic role of BCL2L12.…”

“…It is worth mentioning that an association between BCL2L12 and BCL2 mRNA expression has been noticed in breast tumors (45). In addition, we previously reported that expression of BCL2L12-A, an alternative transcript of BCL2L12, appears to be of importance in colon cancer, since it is associated with Dukes' stage and lymph node status (49). Remarkable modulations of BCL2L12 mRNA levels have also been observed in HL-60 (50-54) and SHI-1 (55) leukemia cells, MCF7 breast adenocarcinoma cells (56,57), OVCAR-3 ovary adenocarcinoma cells (58), AGS (59) and SGC7901 (60) gastric cancer cells, after treatment with various antineoplastic agents.…”

“…Hence, novel prognostic markers are needed to stratify patients into treatment groups. Analysis of BCL2L12 mRNA expression revealed that both splicing variants of this apoptosis-related gene are overexpressed in colon cancer samples as compared to their paired normal mucosa [46,47]. Furthermore, expression of BCL2L12-A, the alternative transcript of BCL2L12, is associated with Dukes' stage and lymph node status [46], whereas higher expression of the full-length BCL2L12 variant is associated with less aggressive forms of intestinal cancer [47].…”
