Expression and function of serum amyloid A, a major acute-phase protein, in normal and disease states

Abstract: Serum amyloid A (SAA), the precursor protein in inflammation-associated reactive amyloidosis (AA-type), is an acute phase reactant whose level in the blood increases in response to various insults. It is expressed in the liver, but its physiological role is not well understood. Recently, a broader view of SAA expression and function has been emerging. Expression studies show local production of SAA proteins in histologically normal, atherosclerotic, Alzheimer, inflammatory, and tumor tissues. Binding sites in … Show more

“…Elevated SAA serum levels have been observed in different tumors, including melanoma,17, 18, 19 which may be due to the inflammatory reactions and tumor necrosis associated with malignant disease. Moreover, SAA protein has also been seen to be expressed by some tumor cells, suggesting that it is directly involved in tumor progression 20. In contrast to our results, SAA has been proposed to be a serum prognostic factor for early‐stage melanoma patients,17 a discrepancy that could be due to a different model design (a different set of variables were considered to perform the analysis) together with a different classification of tumor stages.…”

“…The role of SAA in the repair processes after tissue damage, detoxification of LPS [31] and down-regulation of the proinflammatory responses [30] is supported by our findings that the increase in milk and serum SAA occurred after clinical signs decreased. At the time of the most severe signs, the SAA concentration had reached only one-fourth of the highest value.…”

“…They may be involved in cholesterol transport and in the early response to injury. The SAAs are precursors of amyloid A, a constituent of the amyloid fibrils that are involved in AA amyloidosis [29,30]. This form of amyloidosis is a complication that is associated with many inflammatory conditions (including rheumatoid arthritis and neoplasia).…”
