Expression and function of the small-conductance Ca2+-activated K+ channel is decreased in urinary bladder smooth muscle cells from female guinea pig with partial bladder outlet obstruction

Abstract: The attenuated expression and function of SK channels, which results in the increased UBSM cells excitability and contributes to DO, was discovered in PBOO UBSM cells, suggesting that SK channels might be potential therapeutic targets for the control of OAB.

“…SK channels have a more dominant role in regulating DSM excitability, such that apamin abolishes the fast hyperpolarizations [ 20 , 39 , 82 , 83 , 84 , 85 ]. For I SK channel modeling, the parameters are adopted from guinea-pig urinary bladder smooth muscle [ 40 ].…”

“…In part, these uncertainties stem from the fact that any particular K + channel blocker may influence the permeability of not just one but two or more of the K + conductances present. According to Soder et al 2013 [ 84 ], Parajuli et al 2012 [ 71 ] and Li et al 2017 [ 39 ], elevated conductance of SK channels hyperpolarizes the RMP of DSM cells. Likewise, recent documents ascribed the hyperpolarization in murine DSM cells to SK channels, although the latter were postulated to be present in neighboring electrically connected interstitial cells, not in the DSM itself.…”

“…We investigated if UCL 1684 relaxation of smooth muscle also acted on other known cholinergic responsive (bladder) or cholinergic non-responsive (vascular) smooth muscle (Herrera and Nelson, 2002;Thorneloe et al, 2008;Li et al, 2017). Pretreatment with UCL 1684 partially inhibited relaxation of bladder contraction ( Figure 4A, top panel, summarized in 4B), and caused little inhibition of phenylephrine-evoked contraction of vascular (aortic) rings (summarized in Figure 4B).…”

“…Moreover, OAB is associated with increased DSM excitability, contractility and spontaneous phasic contractions. Therefore, changes in the expression and function of DSM regulatory proteins may alter the contractility of the DSM [1214].…”
