Expression and Subcellular Localization of p42IP4/ Centaurin‐α, a Brain‐specific, High‐affinity Receptor for lnositol 1,3,4,5‐Tetrakisphosphate and Phosphatidylinositol 3,4,5‐Trisphosphate in Rat Brain

Abstract: Recently emerging evidence suggests important roles for inositol polyphosphates and inositol phospholipids in neuronal Ca2+ signalling, membrane vesicle trafficking and cytoskeletal rearrangement. A prerequisite for a detailed physiological characterization of the signalling of both potential second messengers inositol-(1,3,4,5)-tetrakisphosphate (InsP4) and phosphatidylinositol-3,4,5-trisphosphate (PtdInsP3) in the nervous system is the precise cellular localization of their receptors. Based on the cDNA seque… Show more

“…Centaurin ␣1 expressed in the adult is also associated with the synaptosome fraction. This result is in agreement with immunohistochemistry results that show centaurin-␣1/p42IP4 localizes to synaptic regions in neuronal sections (Kreutz et al, 1997;Sedehizade et al, 2002) and is consistent with the identification of centaurin ␣1 in the post-synaptic density fraction by proteomic analysis (Yoshimura et al, 2004).…”

“…1B). Recognition by this antiserum confirms unequivocally the identity of the recombinant protein as we have demonstrated previously that the antiserum stains p42 IP4 specifically in Western blots [31] and also in immunohistochemistry of brain sections [35]. The recombinant GST±p42 IP4 fusion protein was also detectable by a rabbit polyclonal anti-GST antiserum (Fig.…”

“…Moreover, CKI␣ has been found to co-localize in neurones with synaptic vesicle markers and phosphorylates some vesicle synaptic associated proteins (21). Interestingly, centaurin-␣ 1 has been shown to associate with presynaptic vesicular structures (53). An actin-associated protein kinase shown to be a member of the CKI family phosphorylates actin in vitro (54).…”
