Expression in Sf9 insect cells, purification and functional reconstitution of the human proton-coupled folate transporter (PCFT, SLC46A1)

Abstract: The proton-coupled folate transporter (PCFT) provides an essential uptake route for the vitamin folic acid (B9) in mammals. In addition, it is currently of high interest for targeting chemotherapeutic agents to tumors due to the increased folic acid requirement of rapidly dividing tumor cells as well as the upregulated PCFT expression in several tumors. To understand its function, determination of its atomic structure and molecular mechanism of transport are essential goals that require large amounts of functi… Show more

“…The presentation by M. Jansen on the “Path to structural understanding of folate transport through PCFT” was aimed to understand PCFT function and determination of its atomic structure and molecular mechanism of transport. These approaches require large amounts of functional protein and several high-level heterologous expression systems were used for human PCFT including insect cells and yeast [ 14 ]. PCFT was functional in whole cell assays in both expression systems and after solubilization, purification to homogeneity, and reconstitution into liposomes.…”

“…With the increasing demand for recombinant proteins, and continued discovery of novel medically useful proteins, the improvement of production technology has become a focus of research exploration. Various excellent expression systems based on different hosts have gradually become the primary tools for protein production, including plant [ 1 ], Escherichia coli ( E. coli ) [ 2 ], insect [ 3 ], yeast [ 4 ], and mammals [ 5 ].…”

“…PCFT was identified as the main pathway for dietary folate uptake in the proximal small intestine in 2006 [ 5 ]. Detailed studies of human PCFT using PCFT constructs modified with amino acids engineered at specific positions, including the use of the substituted cysteine accessibly method (SCAM) [ 57 ] as well as recent cryo-EM structures of chicken PCFT have provided insights into the topology and conformational transitions during substrate translocation [ 15 , 17 , 33 40 , 45 , 58 61 ]. However, much remains unknown about PCFT’s detailed molecular structure, substrate translocation mechanism, and lipid dependence.…”
