Expression of bone morphogenetic protein-2 and its receptors in epithelial ovarian cancer and their influence on the prognosis of ovarian cancer patients

Abstract: BackgroundTo determine the expression of bone morphogenetic protein-2 (BMP-2) and its receptors BMPRIA, BMPRIB, and BMPRII in epithelial ovarian cancer (EOC) and to analyze their influence on the prognosis of ovarian cancer patients.MethodsSemi-quantitative RT-PCR and western blot were applied to detect the expression of BMP-2 and its receptors BMPRIA, BMPRIB, and BMPRII in EOC, benign ovarian tumors, and normal ovarian tissue at the mRNA and protein levels. Immunohistochemistry was used to determine the expre… Show more

“…The prognostic impact of BMP-2 levels in cancer patients has been evaluated in some studies (Park et al, 2008;Ma et al, 2010). Using immunohistochemical analysis, Ma et al evaluated the influence of intratumoral BMP-2 on five-year survival in 100 patients with epithelial ovarian cancer, showing that the median five-year survival was significantly longer in the groups with a low intratumoral BMP-2 staining intensity (40.32%) than that in the group with tumors expressing high levels of BMP-2 (18.42%, P=0.023) .…”

“…Both normal human ovarian surface epithelial (OSE) cells and EOC cells possess the signalling components necessary for activation of this pathway in response to ligands of this superfamily, including TGF-b, BMPs, activin and Mullerian inhibiting substance (MIS) [6][7][8][9][10][11][12][13]. For example, EOC cells respond to exogenous TGF-b by inducing growth arrest due to upregulation of p15 expression [14].…”

“…The two grades were added, and the specimens were assigned to four levels: 0-1 score (-), 2 scores (+), 3-4 scores (++), and more than 5 scores (+++). The sum of the number of (++) and (+++) cells divided by the total cell number was used to express the percentage representative of the rate of positive expression [25]. CD34-labeled microvessel density (MVD) was quantified based on the methods established by Weidner et al [26].…”
