Expression of galectin‐3 in neuronally differentiating PC12 cells is regulated both via Ras/MAPK‐dependent and –independent signalling pathways

Abstract: Galectin-3 (gal-3) is a member of the galectin family of lectins whose expression strongly depends on the cellular state. Here we show that in PC12 cells the expression of gal-3 protein is regulated via Ras-and mitogen-activated protein kinase (MAPK)-dependent and -independent signalling pathways and correlates with nerve growth factor (NGF)-mediated neuronal differentiation. Gal-3 expression, activation of the MAPK ERK1/2 and neurite outgrowth are induced by NGF and basic fibroblast growth factor (bFGF), but … Show more

“…Furthermore, gal-1-null mice show neuronal abnormalities in adulthood (25). Together with the role of gal-1 in angiogenesis described here, these data strongly suggest that gal-1, as well as other members of the galectin family [galectin-3 (29,30)], is also involved in both neuronal and vascular development.…”

“…For example, the expression of galectin-3 correlates with nerve growth factor (NGF)-mediated neuronal differentiation in PC12 cells and is regulated via Ras-and mitogenactivated protein kinase (MAPK)-dependent and independent signaling. 26 Expression of galectin-3 in skeletal tissues is controlled by Runx2, the transcription factor which regulates osteoblast development and chondrocyte differentiation in the growth plate. 27 Taken together, the expression of galectin-3 appears to be tightly controlled in particular by NF-κB.…”

“…RNA extraction, cDNA preparation, and generation of homologous exogenous competitors for cRT-PCR were performed essentially as described (Kuklinski et al, 2003). Briefly, total RNA reverse transcribed by using the Superscript II Preamplification System (Invitrogen) and random hexamers.…”

“…Cell extractions were carried out as described (Kuklinski et al, 2003). Briefly, cells were solubilized for 30 min at 48C in 50 mM NaH 2 PO 4 /Na 2 HPO 4 , 100 mM NaCl, 0.2 mM CaCl 2 , 0.2 mM MgCl 2 , 1% Triton X-100, pH 7.4, containing the protease inhibitors 4-(2-aminoethyl) benzenesulfonyl fluoride, aprotinin, bestatin, E-64, iodoacetamide, leupeptin, pepstatinA, phenylmethylsulfonylfluoride, and soybean trypsin inhibitor.…”
