Expression of Interleukin 2 Receptors in T Cells Transformed by Strains of Herpesvirus saimiri Representing Three DNA Subgroups

Abstract: Eight T lymphoblastoid cell lines transformed by Herpesvirus saimiri were studied for requirement of interleukin 2 (IL-2) for growth in tissue culture and expression of the IL-2 receptor. The results show that four cell lines grew independent of IL-2, but four other cell lines required IL-2 for optimal growth. However, all H. saimiri transformed cells expressed IL-2 receptors, regardless of IL-2 requirement. Between 2,250 and 8,800 high-affinity receptor sites/cell were detected by ligand-binding assays; in co… Show more

“…HVS strains are classified into three subgroups, A, B, and C, on the basis of their transforming potential and differences in the terminal long unique DNA sequence (49,50). Among these subgroups, only subgroups A and C have been shown to immortalize T lymphocytes of marmosets and other New World primates (21).…”

“…In monkeys and rabbits the virus transforms CD8 + lymphocytes (Kiyotaki et al, 1986) and causes malignant lymphoproliferations (Daniel et al, 1974;Melendez et al, 1969). Several data indicate that lymphokine and growth factor upregulation is involved in the process and mediate autocrine cell growth (Medveczky & Medveczky, 1989). High-level interleukin (IL)-2 receptor expression (Medveczky & Medveczky, 1989) and IL-4 secretion was detected with transformed monkey cell lines (M. Medveczky, P. Geck, C.-S. Chou, A.…”

“…Both viruses can induce a similar pathology after infection of rabbits. The lymphocytes involved are of an analogous phenotype, and lymphocytic cell lines can be established from naturally and experimentally infected animals (26,36,45,72).…”
