Expression of matrix metalloproteinases 1, 3, and 9 in differing extents of tendon retraction in the torn rotator cuff

Abstract: Elevated expression of MMPs 1 and 9 as well as decreased MMP 3 expression can be detected in torn rotator cuff tendon tissue. There is a significant association between the extent of tendon retraction and MMP 9 expression. The results of this study give evidence that early surgical treatment of small and partial-thickness rotator cuff tears is required.

“…Extracellular matrix can also be affected by the activity of metalloproteinases (MMPs), and signifi cant expression of the MMP9 has been detected during rotator cuff and Achilles tendon tears (Karousou et al, 2008;Lakemeier et al, 2011). Moreover MMP-9 was the gelatinase required for the removal of denatured collagen fragments and mediated by the vascular invasion of hypertrophic cartilage callus in developing murine long bones (Colnot et al, 2003).…”

“…TNFα and IL-1β are the upstream genes of MMP, COX-2, IL-6 and IL-17A. Accumulation of MMP and COX-2 induces secretion of SDF-1α, which further promotes inflammation and angiogenesis (32). TGFβ promoted GC cell proliferation, invasion and metastasis (33,34).…”

“…This different role appears to be associated with tendon retraction following a tear in the rotator cuff, since an elevated expression and enzymatic activity can be detected for MMP-1 and MMP-9. This evidence appears to be associated to the extent of the tendon retraction, being accompanied by a marked decrease of MMP-3 activity compared to healthy tendons [101]. Therefore, a tendinopathy seems to be characterized by an imbalance between collagen fibril cleavages by some MMPs, such as collagenases and gelatinases, and a "failed healing response", due to reduced stromelysin activity.…”
