Expression of matrix metalloproteinases 2 and 9 and tissue inhibitors of matrix metalloproteinases 2 and 1 in the glomeruli of human glomerular diseases: the results of studies using immunofluorescence, in situ hybridization, and immunoelectron microscopy

Abstract: The study results suggest that the expression levels and patterns of MMPs and TIMPs are generally similar in several types of glomerular diseases, even though each case has a somewhat different distribution and intensity of expression. When these enzymes were present, their main sites were as follows: MMP-2 was found along glomerular basement membrane, TIMP-2 was located in the acellular mesangial area, MMP-9 was seen in the mesangium, and TIMP-1 was hardly detected. MMP-2 expression is clearly demonstrated to… Show more

“…Meanwhile, the elevated TGF-β1 levels induced the secretion of MMP2 and TIMP2, suggesting that there may be some relationship among TGF-β1 expression, ECM accumulation, alteration of the MMPs/TIMPs system and the renoprotective effects of berberine (Sekiuchi et al, 2012). However, the relationships and mechanisms remain incompletely understood.…”

“…In a megakaryoblastic cell line, rsCD154 increases MMP-9 while not changing MMP-2 expression [Schonbeck et al, 1997;Fernandez Bello et al, 2012]. Moreover, MMPs/TIMPs imbalance have been detected in various glomerulopathies [Catania et al, 2007;Sekiuchi et al, 2012].Defects in either podocyte functions or in GBM composition can lead to proteinuria [Tryggvason and Wartiovaara, 2001;Jarad et al, 2006;Suh and Miner, 2013]. The deleterious consequences of increased MMP-9 expression may stem from both origins.…”

“…Expression of various TIMP family members has been reported in kidney cells, including glomerular mesangial and epithelial cells, tubulointerstitial cells, and cystic cells in culture (4,13,34,50,57,71,73). Expression has also been reported in proximal tubule cells, but primarily only in response to insults, mitogens, or second messengers (14,20,41,45,62,63).…”
