Expression Pattern of Apoptotic Genes in Vitrified-Thawed Bovine Oocytes

Abstract: This study describes a method for quantification of transcripts from low numbers of bovine oocytes using real time RT-PCR. The objective was to evaluate the expression pattern of apoptotic genes (Fas, FasL, Bax and Bcl-2) in vitrified-thawed oocytes. Oocytes were evaluated at germinal vesicle stage; at 15 h of maturation; after vitrification and warming at 15 h of maturation and at 9 h of additional maturation. All transcripts showed an increase in at least 1.2-fold change post-vitrification warming, but the l… Show more

“…In agreement with mentioned results, some researchers showed that the expression of apoptotic genes in the follicles derived from vitrified-warmed ovarian tissue strips were similar to the control group and no sign of ultrastructurally apoptosis were seen in the vitrified follicles [45]. Additionally, Anchamparuthy et al [46] demonstrated that transcript levels of Fas, FasL, Bax and Bcl-2 increased post vitrification-warming and maturation of germinal vesicle oocytes but despite of their prospect any evidence of related apoptosis were not observed in embryos derived from large numbers of vitrified oocytes.…”

“…Vitrification does not alter the expression pattern of BAX in canine oocytes or mouse embryos [90,103], whereas Bcl2 is strongly expressed in vitrified-warmed oocytes [90]. In contrast, vitrification has been reported to up-regulate pro-apoptotic genes (Fas, FasL, Bax, and Bcl-2) in bovine oocytes [93] and down-regulate Bcl2 in mouse embryos compared with that in the control [104]. CD9 is a four-transmembrane superfamily protein located on the plasma membrane of the mouse oocyte [105], and is essential for gamete fusion [106].…”
