Expression pattern of the mouse T gene and its role in mesoderm formation

Abstract: Formation of mesoderm is a crucial event in vertebrate development, establishing many of the important features of the body. Recent studies have implicated molecules that are similar to growth factors in mesoderm formation in Xenopus, but other gene products involved in this process have yet to be identified. Genetic evidence indicates that in the mouse the T gene (Brachyury) has a role in the formation and organization of mesoderm. Mice homozygous for mutant alleles of the T gene do not generate enough mesode… Show more

“…The transcription factor brachyury (T ) marked the posterior epiblast and the nascent primitive streak both in control and N1ICD epi littermates (n=3; Fig. 5G,G′) (Wilkinson et al, 1990). Cer1, encoding a NODAL antagonist expressed in AVE cells at the anterior pole of the embryo (Biben et al, 1998), was expressed normally in N1ICD epi embryos (n=4; Fig.…”

“…In brief, embryos were dissected and fixed overnight in 4% paraformaldehyde (PFA). The following probes were used: Nodal (Conlon et al, 1994), Cerberos-1 (Cer-1) (Thomas et al, 1997), Brachyury (T) (Wilkinson et al, 1990), Hex (Thomas et al, 1998), Sonic hedgehog (Shh) (Echelard et al, 1993), Lim1 (kind gift from M. Jakobson). For generating Bmp-4 probe primers, Bmp4-F 5 0 -TGATACCTGAGACGGGAAG-3 0 and Bmp4-R 5 0 -AACGATCGGCTGATTCT GAC-3 0 were used.…”
