Extension of the diffraction resolution of crystals

Abstract: The diffraction resolution of crystals of the guanine nucleotide-exchange factor complex, EF-Tu-Ts, has been extended from 5.0 to 2.5 A by lowering the solvent content in the crystals as well as the temperature of data collection. The common form of EF-Tu-Ts crystal belongs to space group P212121 with a = 81.1, b = 109.9, c = 207.5 A and has a solvent content of 61%. The crystals diffract to a resolution of 5.0 A, at 293 K and 4.0 A at 273 K. When cryoprotective agents are slowly diffused into the crystals, th… Show more

“…Two reports have appeared in which increased diffractivity is produced by a solute-replacement protocol (Yennawar, Yennawar & Farber, 1994;Schick & Jurnak, 1994). A common denominator of these and the present report is a decrease in the activity of water in the external solution and thus within the crystal.…”

“…Further, post-crystallization methods to enhance crystal quality-dehydration, annealing, soaking, and other treatments-became popular and cured many "poor" crystals (Heras and Martin, 2005). Here as well, observations on crystals of proteins from the translation machinery-a GlnRS and EF-Tu-where among the first that opened the field (Rould et al, 1991;Schick and Jurnak, 1994). Second, the access to strong synchrotron X-ray light sources increased sharply with the building of new third generation facilities worldwide.…”

“…In addition, sometimes the soak in the cryo-solution, especially PEGs, causes dehydration which results in increased order (e.gs. Schick & Jurnak, 1994;Esnouf et al, 1998).…”
