External Quality Assessment for Enterovirus 71 and Coxsackievirus A16 Detection by Reverse Transcription-PCR Using Armored RNA as a Virus Surrogate

Abstract: Three armored RNAs (virus-like particles [VLPs]) containing target sequences from enterovirus 71 (EV71) and coxsackievirus A16 (CA16) and a pan-enterovirus (pan-EV) sequence were constructed and used in an external quality assessment (EQA) to determine the performance of laboratories in the detection of EV71 and CA16. The EQA panel, which consisted of 20 samples, including 14 positive samples with different concentrations of EV and either EV71 or CA16 armored RNAs, 2 samples with all 3 armored RNAs, and 4 nega… Show more

“…Since these initial results, Armored RNA has been applied as a control for a vast array of assays, which include the detection of HCV, 404-406 respiratory viruses such as influenza and severe acute respiratory syndrome (SARS) viruses, 407-409 enteroviruses, 410, 411 and West Nile virus. 412 It has even been utilized as a surrogate virus for the detection of animal pathogens, including classical swine fever virus (CSFV), FMDV, and vesicular stomatitis virus (VSV).…”

“…63,64 To ensure the comparability of results from different laboratories, the external quality assessment for EV71 and CA16 detection using RT-PCR has been established. 65 In addition, a reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay has been developed for sensitive and accurate detection of CA16 and EV71 infections. …”

“…Moreover, MS2 PLP can be stored for a long time without degradation (WalkerPeach et al, 1999; Beld et al, 2004; Hietala and Crossley, 2006; Wei Y.X. et al, 2008; Yu et al, 2008; Zhan et al, 2009; Song et al, 2011). Another positive feature of MS2 PLP is that they are not infectious and thus working with them is safe for laboratory personnel.…”
