“…The blocks were fixed by immersion in SainteMarie's fixative (99% absolute ethanol and 1% glacial acetic acid) overnight (Sainte-Marie, 1962;Tuckett et al, 1988). This alcohol-based fixative was selected to minimize the loss of HA, which is a water-soluble polysaccharide, and we also obtained good results when using the Sainte-Marie's fixative in histochemistry of HA (Felszeghy et al, 2000;Matesz et al, 2005). The blocks were embedded in paraffin, and sections were cut with a Reichert microtome at a thickness of 10 m. After deparaffination, the sections were treated with 2% hydrogen peroxide in phosphate-buffered saline (PBS), pH 7.4, for 30 minutes and with 1% bovine serum albumin (BSA) in PBS for 30 minutes.…”
