Extraction of bioactive compounds against cardiovascular diseases from Lentinula edodes using a sequential extraction method

Abstract: Three extraction methods were sequentially combined to obtain fractions from Lentinula edodes (shiitake mushrooms) containing bioactive compounds against cardiovascular diseases (CVDs). Fruiting bodies were first extracted with plain water, obtained residue was then submitted to supercritical fluid extraction (SFE) and remaining residue submitted to hot water extraction. Sequential design allowed reutilization of the nonextracted material as raw material for the successive extractions increasing extraction yie… Show more

“…Indeed, these extraction strategies have resulted in higher hit leads during drug discovery [ 12 , 38 , 39 , 40 ]. Innovative extraction technologies including semi-bionic extraction [ 41 ], supercritical fluid extraction [ 42 , 43 , 44 ], microwave-assisted, ultrasonic-assisted and enzyme-assisted extraction [ 45 ], molecular distillation methods [ 46 , 47 ] and membrane separation technology [ 48 , 49 ] can be used to extract natural compounds efficiently from plants. These extractions strategies have been shown to have similar simulation to traditional methods allowing the extraction process to get most compounds from the natural product.…”

“…These mostly constitute compounds belonging to secondary metabolic pathways with prominent examples including taxol from Taxus brevifolia, vinblastine from Catharanthus roseus, doxorubicin from Streptomyces peucetius, and cyclosporine from Tolypocladium inflatum [3,4]. The first step in the discovery of lead compounds from natural sources is the release of bioactive metabolites from their biomass through various extraction techniques viz., supercritical fluid extraction [5,6], microwave-assisted and ultrasonic-assisted extraction [7], molecular distillation methods [8], and membrane separation technology [9]. Moreover, bioassay-guided fractionation using chromatographic methods such as preparative high performance liquid chromatography (HPLC) is applied for the isolation and purification of active metabolites from their crude extracts [10].…”

“…These conventional methods rarely produce a significant quantity of the active compound. The innovative technologies that are based on supercritical fluid extraction [39,40], microwave or ultrasound power [41], or membrane separation technology [42] can significantly help to overcome the disadvantages of classical methods. Other nonconventional techniques, such as electrotechnologies (high voltage electric discharge or pulsed electric field), are promising tools for the isolation of bioactive compounds from plant material [38,43] ( Figure 1).…”
