Extraction, selenylation modification and antitumor activity of the glucan from Castanea mollissima Blume

Abstract: The present study aimed to characterize the glucan from C. mollissima Blume fruits and its selenium derivative, then investigate their antitumor activity in vitro. A glucan, designated as CPA, was firstly isolated from the fruits of C. mollissima Blume. Structure analysis indicated that CPA was a linear 1,6-α-D-glucan with the average molecular weight about 2.0 × 10 kDa. The selenylation modification derivative of CPA (sCPA), exhibited a stronger antiproliferative effect on tumor cells than CPA in vitro. CPA a… Show more

“…Proposed structure of Se‐containing polysaccharide generated by the nitric acid–sodium selenite (HNO 3 ‐Na 2 SeO 3 ) method with 1‐6‐linked polysaccharide. Selenylation with ‐HSeO 3 group can occur at C‐2, C‐3, and C‐4 positions and at C‐1 position of the reducing sugar (H. Li et al., 2017). …”

“…And water decoction and ethanol precipitation method is the most common technique for extracting Se-polysaccharides from biological matrix. The method usually exhibited the following advantages: (1) low solvent consumption, (2) low cost, and (3) operating simplicity [ 28 , 29 ]. Ultrasonic extraction was also used to isolate Se-polysaccharides from Se-enriched Lyophyllum decastes .…”

“…Moreover, its operation is simpler than the second method [ 39 ], although it is more time-consuming. For instance, selenylation of glucan from Castanea mollissima fruits with Se content of 574 μg/g was performed using NA-SS system at 65 °C for 10 h [ 29 ]. Likely, selenylation of polysaccharides from Capparis spinosa with Se content of 2438–5545 μg/g was conducted using HA-SS system at 65 °C for 10 h [ 40 ].…”
