Facilitation of estrous behavior by vaginal cervical stimulation in female rats involves α1-adrenergic receptor activation of the nitric oxide pathway

Abstract: In estrogen-primed female rats, vaginal cervical stimulation (VCS) provided by male intromissions or by an experimenter enhances estrous behaviors exhibited by females during subsequent mating with a male. We tested the hypothesis that alpha(1)-adrenergic receptors, acting via the nitric oxide-cGMP-protein kinase G pathway, mediate VCS-induced facilitation of female reproductive behaviors. Ovariectomized, estradiol-primed rats received intracerebroventricular (icv) infusions of vehicle or pharmacological antag… Show more

“…However, another possibility is that PR acts as a common molecular effector that mediates the facilitatory actions of such progestins on lordosis. This idea is supported by the ability of the PR antagonist RU 486 to attenuate the behavioral effects of reduced progestins when they are administered systemically or intracerebrally [9,10,11,12,13]. …”

“…Two weeks later, they were anesthetized with xylazine (4 mg/kg) and ketamine (80 mg/kg), and placed in a Kopf stereotaxic instrument (Tujunga, Calif., USA) for implantation of a stainless steel cannula (22 g, 17 mm long) into the right lateral ventricle following coordinates from the atlas of Paxinos and Watson [32] (A/P, +0.80 mm; M/L, –1.5 mm; D/V, –3.5mm with respect to bregma); these coordinates had been used in previous works from our group [10]. A stainless steel screw was fixed to the skull and both cannula and screw were attached to the bone with dental cement.…”

“…Vaginocervical stimulation during pro-oesterus/oestrus elicits citrulline production, a marker of NO synthesis, in the MOB (Guevara-Guzman et al, 2000). This effect may depend on noradrenergic activation of the NO/cGMP/protein kinase G pathway (Gonzalez-Flores et al, 2007). Indirect evidence that NO affects olfactory processing in the main olfactory bulb is based on the ability of nNOS blockers applied to the main olfactory bulb to block olfactory memory formation (Kendrick et al, 1997).…”

“…It must be noted that the relationship between creatinine and urea and the actual GFR is hyperbolic rather than linear. Hence, initial elevation of serum concentrations of these substances indicates a large decrease in renal performance (22).…”
