Fate of the first polar bodies in mouse oocytes

Abstract: Both nuclear transfer and intracytoplasmic sperm injection (ICSI) practice necessitates studies on the spatial relationship between the MII spindle and the first polar bodies (FPB). Although recent observations have shown that the FPB position does not predict accurately the location of the meiotic spindle in metaphase II oocytes of monkey, hamster, and human, detailed studies on FPB deviation and its affecting factors are lacking. Since polar bodies can be used for genetic testing and oocyte quality grading, … Show more

“…Wide PVS may be seen because of overmaturity of cytoplasm at the time of hCG injection. So, PVS enlargement may be related to increased maternal age, and oocyte aging [22,35]. One study detected enlarged PVS as a sign of degeneration/postmaturity in unfertilized human oocytes from an ultrastructural point of view [23].…”

“…Procedures for animal handling, superovulation and oocyte collection were those described previously [39,40], with modifications. Briefly, mice of the Kun-ming breed, originally derived from ICR (CD-1), were kept in a room with 14 h/10 h light-dark cycles, the dark starting from 8 PM.…”

“…In addition, different batches of FBS were used during the experiments, and the results remained unchanged (data not shown). In mice, PB degeneration has been associated to oocyte's ageing (Miao et al, 2004) and apoptosis (Jiao et al, 2012). In our conditions, FBS …”
