Fatty acids and TxA2 generation, in the absence of platelet-COX-1 activity

Abstract: Background and aims Omega-3 fatty acids suppress Thromboxane A2 (TxA2) generation via mechanisms independent to that of aspirin therapy. We sought to evaluate whether baseline omega-3 fatty acid levels influence arachidonic acid proven platelet-cyclooxygenase-1 (COX-1) independent TxA2 generation (TxA2 generation despite adequate aspirin use). Methods and results Subjects with acute myocardial infarction, stable CVD or at high risk for CVD, on adequate aspirin therapy were included in this study. Adequate as… Show more

“…Omega-3 fatty acids also exert antithrombotic effects by various mechanisms including a reduction in thromboxane synthesis [ 99 ] and PAF [ 100 , 101 ]. Omega-3 fatty acids are incorporated in the cell membrane and may regulate the activity of PLA 2 and thus lyso-PAF production, which is a prodrome molecule for PAF production [ 102 ].…”

“…Of the 21 studies, ten were included in the meta-analyses [ 10 , 14 , [18] , [19] , [20] , [21] , [22] , [23] , [24] , [25] ]. The other 11 studies were not included due to having incomplete information [ 26 , 27 ], results given in different units [ [28] , [29] , [30] , [31] ], study results had been published in another study [ 1 , 32 ], measurements were reported from plasma concentrations [ 13 ] or included diseased populations [ 33 , 34 ]. Characteristics of all studies included in the analyses can be found in Table 1 .…”

“…Moreover, ω-3 PUFAs induce the suppression of thromboxane A 2 (a factor responsible for platelet aggregation, vasoconstriction and fibrinogen reduction) synthesis, and favor the synthesis of thromboxane A 3 [64][65][66]. In this context, animal models highlighted that EPA consumption also plays a role in stabilizing the atheromatous plaque [67].…”
