Fatty Liver and Insulin Resistance in the Liver-Specific Knockout Mice of Mitogen Inducible Gene-6

Abstract: Mitogen inducible gene-6 (Mig-6) is a feedback inhibitor of epidermal growth factor receptor (EGFR) signaling pathway. The liver-specific knockout mice of the Mig-6 gene (Mig-6d/d) showed hepatomegaly and increased hypercholesterolemia. In this study, the biomarkers of insulin resistance and the effects of high-fat diets in the wild (Mig-6f/f) and Mig-6d/d mice were analyzed. The fasting plasma concentrations of glucose, triglyceride, cholesterols, free fatty acids, and HOMA-IR were measured and the glucose to… Show more

“…Moreover, it has been reported that ERRFI1 regulates cholesterol and glucose metabolism in the liver. Errfi1 liver-specific knockout in mice altered expression of lipid and glucose metabolism-related genes, resulting in increased blood cholesterol and fasting glucose levels and leading to insulin resistance [71][72][73]. In Fig.…”

“…Hu et al [30] found that Gosha-jinki-gan ( GJG ), composed of 10 herb CHM, is not only useful for DKD but also can improve the glucose utilization and insulin resistance in STZ-induced diabetic rats. Lipid deposition can also lead to insulin resistance and restricted the use of glucose capacity in the muscle [31]. Wen-Pi Tang , which is composed of five CHM ( Rhei Rhizoma , Ginseng Radix , Aconiti Tuber , Zingiberis Rhizoma , and Glycyrrhizae Radix ), showed its protective effects by virtue of the reversal metabolic abnormalities associated with hyperglycemia and hyperlipidemia [32].…”

“…It is particularly important for diabetic patients to actively prevent dyslipidemia. Lipid deposition in the liver causes liver insulin resistance (Park et al, 2016 ). Lipotoxicity increased islet cell apoptosis (Huang C. N. et al, 2017 ) and restricted the use of glucose capacity in muscle.…”
